I have no experience with locking tuners. Is it worth upgrading from standard Jackson tuners to Gotoh locking tuners, if you're already using a Floyd Rose 1000 with locking nut?

I am not a proponent of using locking tuners with a floyd. it does not buy additional tuning stability. it has been claimed by others that the locking tuners make changing strings faster, but I have not necessarily found this to be the case to any significant degree. yes, you can use no wraps, but there's additional steps to lock each string. even with non-locking tuners, when using a locking nut you don't need a ton of wraps on the post.
this is personal taste but I also think tuners with thumbwheels on the back look clunky, as opposed to the sleek design of the non-locking kind. gotoh makes (or used to make) these "self-locking" tuners which don't have a thumbwheel. they work well, and I have a set on one of my hardtail guitars, but again, wouldn't necessarily recommend for a floyded guitar.

I agree with metalhobo. All of the tuning stability comes from the locking nut, so locking tuners won't do you anything on a floyd.
I string my floyds with the string "backwards". I put the ball end on the tuner side, then cut to size to put them in the floyd block. Quick and easy, you only have to cut once, and you don't have sharp ends on the headstock.
